Protein folding and misfolding are two halves of the same coin. The earlier protein-folding conundrum explores how a protein’s amino acid sequence dictates its 3D structure. Over fifty years of studying the equilibrium thermodynamic and kinetic mechanisms of folding have produced both computational methods and artificial intelligence programs that are able to design non-biological polymers and proteins, as well as predict native protein structures with relatively high accuracy.
The sibling field of protein misfolding, in parallel, has also been rapidly developing. Proteins that fail to fold into their proper native structures assemble into fibrillar amyloid aggregates, which are seen in diseases such as neurodegeneration. Recent research in liquid–liquid phase separation (LLPS) offers a potential alternative to the fibril formation pathway. Unraveling the mechanisms and consequences of misfolding is imperative to studying and developing treatments for relevant proteinopathies.
In light of the longevity and growth of this field, this Special Issue will include articles that contribute to the recent advances in protein folding and misfolding.

Submitted manuscripts should not have been published previously, nor be under consideration for publication elsewhere (except conference proceedings papers). All manuscripts are thoroughly refereed through a single-blind peer-review process.
