Dear Colleagues,

Blood–brain barrier dysfunction is a pathological hallmark of several inflammatory-associated diseases in the central nervous system, including multiple sclerosis, stroke, dementia, traumatic injuries, encephalopathies, and brain metastases. Loss of BBB integrity leads to increased leukocyte infiltration, plasma protein extravasation, and edema, which may ultimately lead to neuronal dysfunction and degeneration. Mounting evidence suggests that therapeutic approaches preserving endothelial barrier function may prove beneficial during bouts of neuroinflammation, however, there remain limited options for BBB-specific treatments.

This Special Issue of Brain Sciences will explore the current knowledge and innovative concepts regarding preclinical and clinical research into the mechanisms of BBB regulation and inflammation-mediated barrier dysfunction. Authors are invited to submit original research, topic reviews, or case studies that will inform neuroscientists, vascular biologists, and clinicians about the relative roles BBB integrity plays CNS homeostasis and inflammation.

Topics include the following areas: BBB dysfunction in disease, BBB regulation during homeostasis or disease, neuroimaging of BBB integrity, BBB-specific therapeutics, endothelial and mural cell biology, leukocyte–endothelial interactions, and endothelial cell–cell junctional regulation.
