The authors have written an interesting paper based  on the study to assess  the Regulation of Syntaxin 1A activity in Ca2+-triggered release in PC12 cells.

Although some interesting data is provided and the methods are appropriate I felt that the paper lacked sufficient background information to explain the relevance of the study and provide some neurological/neurochemical information. This lack of information makes the study seem quite esoteric which isn`t, but needs more detail. Furthermore, PC12 cells were used in the study therefore, it should be  emphasized that the results apply to rats and not as yet humans. Details about the PC12 cells was available in the methods. Furthermore, some information on how the results influence neurotransmitter release would be appropriate. Overall, a good study, but more information in the introduction required.

This manuscript addresses an important study about CK2 Phosphorylation required for Regulation of Syntaxin 1A activity in Ca2+-triggered release in PC12 cells.

This study represents a massive effort and the proposed mechanism suggested by dynamic FRET analysis is quite convincing.

I believe the paper will be of interest to the readership of IJMS and would recommend it for acceptance.
